My wife and I have played this multiple times as two player and we really enjoy it. We have also played a couple of 5 player games and I am surprised to say we actually enjoyed the two player more. For us it is the fun of completing quests as opposed to "sticking it" to the other player(s), that is why we made a house rule that no mandatory quest can be given on the last round.
I am going to give this game the highest complement I can - it is just plain fun.
BTW: Carcassonne is my wife's favorite game and she really enjoys LoW.
